The doctoral student project aims at generating new knowledge about the impact of small-area deprivation on inequality in health. The student will investigate the spatial distribution of disease burden by small-area deprivation across regions in Sweden and explore if the socioeconomic standard of living areas can impact the health outcomes of its inhabitants. The project will build upon nationwide register data on health, health care and welfare and methods developed in the Global burden of Disease (GBD) study. The goal is to improve the knowledge around the impact of small-area deprivation and inequality in health and to help support policy decision and targeted action.

PhD position summary/title: Doctoral (PhD) student position in Adipocyte Systems Biology

We are seeking a highly motivated and ambitious PhD student to join the research group of Drs. Mikael Rydén (co-supervisor) and Niklas Mejhert (main supervisor) at the Department of Medicine, Huddinge. The project will be performed in collaboration with Dr. Patrik Ståhl (co-supervisor) who leads a laboratory at KTH Royal Institute of Technology which is world-leading in spatial mapping approaches. Together, we will determine how the cellular landscape and microarchitecture of white adipose tissue is affected in health and disease. The studies are based on integrative analyses of single-cell, single-nucleus and spatial transcriptomic data and the results will followed-up in different cell culture systems of human adipocytes. Most of the work will be carried out in the Rydén and Mejhert laboratory in the NEO research building at Karolinska Institutet Campus Flemingsberg.

PhD position summary/title: PhD position in Neuroimaging and Deep Learning in Alzheimer’s Disease

In this research project, the student is expected to apply methods based on deep neural networks to identify the anatomical and functional brain connections that are most vulnerable to Alzheimer’s disease and assess whether they can predict cognitive decline. The student will also have the opportunity to learn and apply computational models to identify the mechanisms responsible for the damage of brain connections in AD. Finally, the student will include the methods developed in this project in the second version of our software (BRAPH), which will soon be released.

The PhD project will examine factors that may prevent adenocarcinoma of the oesophagus, a cancer with increasing incidence and poor prognosis. In large-scale studies, based on merged data from several national health data registries in Nordic countries, the project aims to evaluate if the risk of oesophageal adenocarcinoma is decreased after obesity surgery, antireflux surgery and use of certain potentially anti-carcinogenic medications, i.e. aspirins, statins, and proton pump inhibitors.

PhD position summary/title: Two doctoral (PhD) student positions in translational medicine & chemical biology

Project 1: OGG1 activators and inhibitors in control of inflammatory diseases

With Professor Thomas Helleday as principal supervisor, supported by Christina Kalderén, Team Leader Oliver Mortusewicz and Assistant Professor Maurice Michel in the co-supervision team, this project aims to understand both positive and negative small molecule modulation in DNA repair. In the past, we have developed OGG1 inhibitors and broadly studied the compounds from basic biochemistry and biology to first disease models (Visnes et al. 2018, Science; Visnes et al. 2020; NARTanner et al. 2021, biorxiv). Currently, we are investigating small molecule activators of OGG1, as a novel technology to enhance oxidative DNA damage repair (Michel et al. 2022, unpublished). The prospective doctoral student will perform research to i) understand the precise mode of action of small molecule activators of OGG1, ii) unite research on inhibitors and activators to elucidate the molecular role of OGG1 in promoting transcription, and iii) use OGG1 inhibitors to evaluate therapeutic opportunities in a disease model of rheumatoid arthritis.

Project 2: Understanding and exploiting drug resistance in cancer – targeting SAMHD1

With Assistant Professor Sean Rudd as principal supervisor, supported by Professor Thomas Helleday and Dr Si Min Zhang in the co-supervisory team, this project seeks to understand and exploit drug resistance pathways in cancer cells. Central to this work is our previous research upon the enzyme SAMHD1 which we established as a targetable chemoresistance factor in acute myeloid leukaemia (Herold et al., 2017, Nat MedRudd et al., 2020, EMBO Mol Med). The doctoral student will build upon these findings and broadly aim to define and exploit this role of SAMHD1. This will entail, (i), the systematic characterisation of the interaction of SAMHD1 with nucleoside drugs, (ii) the development and benchmarking of small molecule SAMHD1 inhibitors, and (iii), defining the broader role of SAMHD1 in modulating the efficacy of oncology drugs.

PhD position summary/title: Doctoral (PhD) student position in liquid biopsy biomarkers of pancreatic beta cell damage

This KID-funded Ph.D. project focuses on developing non-invasive methods for identifying individuals at risk of type 1 diabetes and as such will contribute to the introduction of personalized medicines for use in preventing this common and life-threatening disease. More specifically, the candidate will contribute to the discovery of protein biomarkers that predict disease onset. The candidate will be part of a supportive team and in this environment, perform global proteomics analyses of the secretome and extracellular vesicles released from cultured cells. The candidate will also contribute to the identification of biomarkers in whole plasma samples and plasma-derived extracellular vesicles collected from an experimental model of type 1 diabetes. The tasks include learning and performing methods for extracting and characterizing extracellular vesicles including nanoparticle tracking analysis, electron microscopy (EM), methods for identifying secreted and extracellular vesicle associated proteins including advanced mass spectrometry (conducted by SciLifeLab), Western blot, etc. It may also include work with experimental disease models and viruses suspected to play a role in triggering type 1 diabetes. The work is performed in the Flodström-Tullberg group with extra support for the studies involving extracellular vesicles provided by the co-supervisor, Samir el Andaloussi and his team. The student will learn how to handle the necessary bioinformatics tools and software for data analysis (proteomics), with guidance from a member of the main supervisor’s laboratory and NBIS (https://nbis.se).

Estrogen signaling is mediated by estrogen receptors which can be activated or inactivated by ligands, and are ideal candidates for therapeutic targeting.
