Mitochondrial dysfunction is not only associated with metabolic defects but also with aging and autoimmunity. In oxidatively-stressed mitochondria, oligomerization of mitochondrial membrane protein VDAC (voltage-dependent anion channel) forms a macropore that releases mitochondrial DNA (mtDNA) into the cytosol and ultimately, into the plasma (https://science.sciencemag.org/content/366/6472/1531/tab-article-info). Released mtDNA is perceived by our immune system as “foreign” and elicits autoimmunity and inflammation. Inhibiting VDAC oligomerization with a small molecule inhibitor prevents type-I interferon signaling and ameliorates autoimmune diseases such as lupus.
